Abstract:Superoxide dismutase 2 (SOD2) gene is an important component of endogenous antioxidant defense barrier and plays important roles in the prevention of disease, anti-aging, anti-tumor and anti-inflammation. To study the expression and localization of SOD2 gene in breast tissue of normal and clinical mastitis sheep (Ovis aries), Breast tissue of 3 normal and 3 clinical mastitis sheep were selected in this study. The expression of SOD2 mRNA and protein in breast tissue of normal and clinical mastitis sheep were detected by qRT-PCR and Western blot. And the SOD2 protein was located in breast tissue of normal and clinical mastitis sheep by immunohistochemistry (IHC) method. The results showed that the expression of SOD2 mRNA and protein in the breast tissue of clinical mastitis sheep were extremely significantly higher than that of control group (P<0.01). Immunohistochemical staining found that SOD2 positive expression was located in breast epithelial cells of normal and clinical breast tissue. SOD2 positive reaction was stronger in breast tissue of the clinical mastitis sheep. The results of this study show that the expression of SOD2 mRNA and protein in the breast tissue of clinical mastitis sheep are up-regulated, will provide basic data for the further study of the mechanism of the gene and the pathogenesis of sheep mastitis.
