不同生态区烟草叶片基因表达谱的分析

Analysis of Differential Gene Expression Profile for Tobacco (Nicotina tabacum L. cv.) Leaf in Three Different Ecological Zones

摘要 烟叶香气类型受到生态环境和栽培条件的影响,为解析生态环境对烟叶香气风格形成的影响,本研究利用定制的烟草(Nicotina tabacum L. cv.)芯片对种植在河南平顶山(浓香型)、云南省玉溪(清香型)和贵州遵义(中间香型)3个不同生态区的同一烟草品种K326的同一叶位(中部叶)、相同叶龄(60 d)的叶片基因表达谱进行了比较研究,共发现有效差异表达基因920个。通过功能注释和代谢途径分类发现,初生代谢途径中碳水化合物的代谢可能对不同香型风格烟叶的形成起到了较大影响作用,而氮代谢的影响较小。而在次生代谢中,苯丙烷类合成、生物碱合成、萜类代谢途径起到较大的作用。另外,氨基酸的代谢可能也对不同香型风格烟叶的形成起到了重要作用;其中,光合作用中起到重要作用的磷酸烯醇式丙酮酸碳羧化酶(phosphoenolpyruvate carboxylase 1, PPC1)、碳水化合物中淀粉代谢相关的细胞壁转化酶(cell wall invertase 1, CWINV1)、萜类代谢关键酶3-羟-3甲基戊二酰辅酶A还原酶(3-hydroxy-3-methylglutaryl coA reductase, HMGR1)和萜类合成酶(terpene synthase 21, TPS21)、以及生物碱合成相关基因均在河南平顶山烟叶中表达量最高,贵州遵义次之,云南玉溪最低;类胡萝卜素合成关键基因八氢番茄红素合成酶(phytoene synthase, PSY)的表达量在河南平顶山最低,其次是云南玉溪,在贵州遵义表达量最高。而类胡萝卜素降解关键酶9-顺式-环氧类胡萝卜素双氧合酶(nine-cis-epoxycarotenoid dioxygenase, NCED4)基因表达量表现出云南玉溪最高,贵州遵义次之,河南平顶山最低。淀粉降解相关的葡聚糖磷酸化酶(α-glucan phosphorylase, PHS1)基因在云南玉溪表达量最高,河南平顶山次之,贵州遵义最低。结果提示,这些基因的差异表达可能对不同香型风格烟叶的形成起到了重要作用,该研究为进一步采用农艺措施调控提供了理论支撑。

Abstract:Tobacco aroma sytles is affected by the ecological environment and cultivation conditions. In order to investigate the effect of ecological environment factors on tobacco(Nicotina tabacum L. cv.) aroma style formation, we characterized gene expression profiles in leaf of the tobacco cultivar K326, which were grown in three main growth areas in China: Pingdingshan County in Henan Province, where is famous for producing strong arama feature of tobacco leaf, Yuxi County in Yunnan Province, where displays mild aroma style of tobacco leaf, and Zunyi County in Guizhou Province, where is representative for producing aroma style between strong and mild of tobacco leaf. A total of 920 genes were found to express differently in three ecological zones. After pathway analysised by KEGG(kyoto encyclopedia of genes and genomes), our results showed that carbohydrate metabolism might have more contribution to the formation of different arama styles. While the biosynthesis of phenylpropanoids, biosynthesis of alkaloids and metabolism of terpenoids, which belong to the secondary metabolism, might have great influence on the formation of arama features. Besides, amino acid metabolism might also have a certain influence on aroma features' formation. Also, we found the gene expression of phosphoenolpyruvate carboxylase 1 (PPC1) (photosynthesis related), cell wall invertase 1 (CWINV1) (starch biosynthesis related), 3-hydroxy-3-methylglutaryl coA reductase (HMGR1) and terpene synthase 21 (TPS21) (terpenoids metabolism related), and some genes related to alkaloid biosynthesis were the highest in Henan Province and lowest in Yunnan Province. While the phytoene synthase (PSY) (key enzyme involved in carotenoid biosynthesis) expressed the highest level in Guizhou Province and the lowest in Henan Province. For the carotenoid degradation, the nine-cis-epoxycarotenoid dioxygenase (NCED4) showed the highest level in Yunnan and the lowest in Henan Province. As for the starch metabolism, the α-glucan phosphorylase (PHS1) expressed the highest level in Yunnan and the lowest in Guizhou Province. These results showed that the expression levels of these genes might play important roles in the formation of different aroma styles under the different ecological environments,which can provide some clues for the regulation of the formation of different aroma styles by proper agronomic practices in the future.
