山羊CIDEa基因过表达对乳腺脂质合成的影响

摘要 诱导细胞死亡的DNA片段化因子-α样效应因子a (cell-death-inducing DNA fragmentation factor-α-like effector a, CIDEa)是一种脂滴相关蛋白,参与调节脂代谢和维持机体能量平衡。为了进一步探讨CIDEa在奶山羊(Capra hircus)乳腺脂质代谢中的功能,本研究以奶山羊乳腺上皮细胞为模型,通过构建pcDNA3.1-CIDEa过表达载体研究CIDEa基因对乳腺细胞脂质合成的影响。结果发现,乳腺上皮细胞转染pcDNA3.1-CIDEa过表达载体48 h后,CIDEa基因表达量上升了近10倍(P<0.01);甘油三酯(triacylglycerol, TAG)检测和油红O染色结果显示,过表达CIDEa基因后乳腺上皮细胞中甘油三酯的含量显著升高(P<0.05)且脂滴积累增多;实时定量PCR检测结果显示,过表达CIDEa基因后脂代谢相关基因乙酰辅酶A羧化酶α (acetyl-CoA carboxylase alpha, ACACA)、抗原分化簇36 (cluster of differentiation 36, CD36)和固醇调节元件结合蛋白1 (sterol regulatory element binding protein1, SREBP1)的表达量显著升高(P<0.05),二酰基甘油酰基转移酶2 (diacylglycerolacyltransferase2, DGAT2)的表达量极显著升高(P<0.01)。以上结果表明,CIDEa基因能促进乳腺上皮细胞脂质的合成。本研究为深入研究CIDEa基因在奶山羊乳脂代谢调控中的作用提供了理论参考。

Abstract:Cell-death-inducing DNA fragmentation factor-α-like effector a (CIDEa) is a lipid droplet associated protein, which is involved in regulating lipid metabolism and maintaining body energy balance. In order to further explore the function of CIDEa in lipid metabolism in mammary gland of dairy goats (Capra hircus), in this study, dairy goat mammary epithelial cells were used as a model to study the effect of CIDEa gene on lipid synthesis by constructing pcDNA3.1-CIDEa overexpression vector. The results showed that the mRNA expression of CIDEa gene was increased nearly 10-fold after 48 h of transfection with pcDNA3.1-CIDEa in dairy goat mammary epithelial cells (P<0.01). The results of triacylglycerol (TAG) contents assay and Oil Red O staining showed that the accumulation of lipid droplets and the content of triglyceride (P<0.05) in cells were increased after CIDEa overexpression. The results of real-time quantitative PCR showed that CIDEa overexpression significantly increased the expression of acetyl-CoA carboxylase alpha (ACACA)(P<0.05), cluster of differentiation 36 (CD36)(P<0.05), sterol regulatory element binding protein 1 (SREBP1)(P<0.05) and diacylglycerol acyltransferase 2 (DGAT2)(P<0.01).These results indicated that CIDEa gene could promote lipid synthesis in mammary epithelial cells. This study provides a theoretical basis for further studying the role of CIDEa gene in the regulation of milk fat metabolism in dairy goats.
