Tissue Culture and Component Analysis of Pinellia ternata |

Abstract Abstract: Pinellia ternata Breit, a Chinese traditional herb, is facing problems of resource declining and requirement increasing. The segments of leaves and petioles from P. ternata were used as explants to induce small tuber. The highest inducing frequency reached 100% on MS medium supplemented with BA 0.5~2.0 mg/L and NAA 0.2 mg/L. One tuber propagated to 16.9 tubers evenly within 3 weeks on MS medium with BA 2.0 mg/L and NAA 0.2 mg/L.The small tubers could be easily induced to plantlets and transplanted to field. The experiment showed that it was practicable to produce large numbers of high quality plantlets by tissue culture. HPLC was employed to analyze the extracts of 80% methanol, 0.2 mol/L H2SO4 and distilled water from wild and tissue culture tubers. The results showed that the total content of the components tested in the tissue culture tubers were higher than that in the wild, and the main chemical composition were similarity between them.
