Abstract Abstract: Ginseng (Panax ginseng C.A.Mey)hairy roots were induced from the root explants of ginseng by the co-culture of Agrobacterium rhizogenes and ginseng roots with the method of direct inoculation. The content of ginsenosides in ginseng hairy roots after 4 weeks culture basically reached the level of that in 3-year-old cultivated ginseng roots, and the content of single ginsenoside-Rb1 had an evident increase. The result indicated that TL-DNA had a function of influencing the biosynthesis of ginsenosides. Based on the analytical results of RiA4TL-DNA sequence, rolC gene affecting the synthesis of ginsenosides was amplified and cloned from the ginseng hairy roots by PCR method. Comparison with previously published nucleotide sequence showed that the homology was 99.9%.
