Abstract Abstract: Rat hepatocytes were treated with clofibrate, the express levels of PXR and CYP3A1 were determined by Western blot or RT-PCR. The results as follows: Clofibrate markedly increased the expression of PXR, and the induction was abolished by actinomycin D, an inhibitor for mRNA synthesis. No induction of CYP3A1 by clofibrate was tested, PCN alone markedly induced the expression of CYP3A1, co-treatment with clofibrate enhanced the induction. Clofibrate was one of pero瑇isome proliferators, the induction of PXR and CYP3A1 by clofibrate provided a molecular mechanism on drug metabolism.
