Experiment on Cryopreservation of Porcine Oocytes by Using Glass Micropipette |

Abstract The self-made glass micropipette(GMP) was used for cryopreservation of pig oocytes, obtaining good results. The post-thaw survival rate of oocytes was assessed by fluorescein diacetate(FDA) staining. And the results showed that the programmed freezing method and GMP vitrification method respectively resulted in the post-thaw survival rate of 34.5% and 63.3%(P <0.05) in cryopreservation of porcine MⅡstage oocytes. Comparing plastic straw with GMP in vitrification of porcine MⅡ stage oocytes, the survival rates of both groups were 45.0% and 65.9% (P < 0.05), respectively. When GV-stage pig oocytes were cryopreserved by straw programmed freezing method and GMP vitrification method, they yielded survival rates of 30.0% and 59.7% respectively and there was significant difference between them(P < 0.05). The present study shows that GMP method is an efficient freezing method for porcine oocyte cryopreservation.
