Isolation and Sequencing Analysis of the Stigma-specific Promoter from Brassica napus |

Abstract SLR1 gene is a stigma-specific S-locus related gene. Primers derived from the SLR1 promoter sequence of Westar were used to perform PCR analysis of several B. napus varieties, so as to isolate corresponding promoters of self-compatible B. napus. The fragments achieved were cloned and sequenced, and then fused to the GFP gene, replaced the 35S promoter in the plant expressed vector pFGC-eGFP. The vector was transformed to Arabidopsis by agrobacterium-mediated transformation. By sequences analysis and fluorescence detection, we concluded that the SLR1 promoter in self-compatible B. napus had similar structure and function with the self-incompatible B. oleracea. The results would lay a foundation for further study on the S-locus genes in Brassica napus.
